Molecular detection of rice tungro bacilliform virus (RTBV) on weed host in South Sulawesi

Abstract Rice tungro bacilliform Virus (RTBV) is one of the viruses that cause disease in rice plants causing leaf yellow and plant stunting. Some weed species have also been described as potential hosts for RTBV addition to rice. However, cannot be diagnosed weeds only by looking at their morphological symptoms because infected with do not show typical symptoms. Hence this research was conducted detect presence molecularly on several fields South Sulawesi. The polymerase Chain Reaction technique coat protein gene-specific primers used RTBV. A total 29 samples consisting 12 collected from Sidrap, Pinrang, Maros, Gowa were then extracted genomic DNA PCR analysis. results analysis showed tested positive Panicum repens (Lawawoi/Sidrap), Echinochloa crusgalli Cyperus difformis (Lanrang/Sidrap), (Palangga/Gowa), Fimbristilys miliacea Eleusine indica (Lawawoi/Sidrap) which indicated virus fragments 430 bp length all these weeds. Weeds formed 2 clusters. first cluster consisted P.repens E.crusgalli Lawawoi/Sidrap, C.difformis F.miliacea Lanrang/Sidrap, E.indica Palangga/Gowa. At same time, second Lawawoi/Sidrap.
